en اثرات دوگانه تجویز دگزامتازون بر میزان اضطراب و افسردگی در موش های صحرایی نر بالغ Nervous system (others) Others Experimental research article Experimental research article مقدمه: دگزامتازون به عنوان یک گلوکوکورتیکوئید سنتتیک دارای اثرات احتمالی در اضطراب و افسردگی است، زیرا اثر مستقیمی بر محور هیپوتالاموس-هیپوفیز-آدرناا و تداخل با چندین سیستم نوروترانسمیتری از قبیل گابا و گلوتامات دارد. 4 در /5، 2 ، 5 ، 24 ، 04 و 14 mg/kg روش ها: سطوح اضطراب و افسردگی موش ها به ترتیب 44 دقیقه و 14 ساعت پس از تزریا ق زیا ر پوسات ی دگزامتاازون در مقااد یر دستگاه ماز بعلاوه مرتفع و تست شنای اجباری اندازه گیری شد. یافته ها: نتایج حاصل از تست اضطراب، کاهش معنی داری در درصد زمان حضور و درصد تعداد ورود باه بازوهاا ی بااز با ین دگزامتاازون 2 و ساال ین و همچنا ین افازا یش معاااانی داری در درصد زمان سپری شده در بازوهای باز بین دگزامتازون 14 و سالین نشان داد. از نظر فعالیت حرکتی )تعداد کل ورود به بازوهای باز و بسته( کاهش معنی داری بین دگزامتازون 2 و سالین، همچنین تفاوت معنی داری بین دگزامتازون 2 و 14 مشاهده شد. مقایساه سانشش افساردگ ی در زماان با ی تحرکا ی کال افازا یش معنا ی داری با ین دگزامتازون 2 و سالین و همچنین کاهش معنی داری بین دگزامتازون 04 و سالین نشان داد. از نظر تأخیر در توقف اولیه نیز بین دگزامتاازون 2 و 04 تفااوت معنا ی داری مشااهده شد. نتیجه گیری: تزریق دگزامتازون احتمالاً اثرات دوگانه ای دارد، مقادیرکم این دارو اثر اضطراب زایی و افسردگی و مقادیر بالای آن اثرات ضاد اضاطراب ی و ضاد افساردگ ی دارد. Introduction: Dexamethasone is a synthetic glucocorticoid with possible effects on anxiety and depression because it has direct effects on the hypothalamus-pituitary-adrenal (HPA) and interacts with several neurotransmitter systems such as GABA and glutamate. Methods: Levels of anxiety and depression in rats were measured 40 minutes and 24 hours, respectively, after subcutaneous injection of Dexamethasone Sodium-Phosphate (DEX) 0.5, 1, 5, 10, 20, 30 mg/kg (saline 1ml/kg) in the elevated plus maze and forced swimming test. Results: The results of anxiety test showed a significant decrease in the percentage of open arm time and open arm entries in DEX (1 mg/kg) group compared to the saline groups. The percentage of time spent in the open arms significantly increased in DEX (20mg/kg) compared to the saline group. In terms of locomotor activity (total number of open and closed arms), significant decrease was observed in DEX (1 mg/kg) compared to the saline group. Also, a significant difference between DEX 1 and 20 mg/kg groups was observed. Comparative assessment of depression during total immobility time showed significant increase in DEX (1 mg/kg) compared to saline and also a significant decrease was observed in DEX (30 mg/kg) compared to the saline groups. A significant difference in latency to immobility was also detected between DEX 1 and 30 mg/kg groups.
