OTHERS_CITABLE بررسی نقش نیتریک اکساید هیپوکامپی در یادگیری احترازی غیر فعال چکیده: مقدمه: نیتریک اکساید یکی از میانجی های عصبی مهم در ناحیه هیپوکامپ است که به صورت یک پیامبر برگشتی در روندهای شکل پذیری سیناپسی و در نتیجه یادگیری و حافظه دخیل می باشد. مطالعات گذشته حاکی از ساخته شدن نیتریک اکساید در نورونهای پیرامیدال هیپوکامپی است. آنزیم سازنده نیتریک اکساید (نیتریک اکساید سنتاز) میتواند هدف فارماکولوژیکی مناسبی برای بررسی نقش نیتریک اکساید در روندهای مختلف شناختی در مغز قرار بگیرد. در این تحقیق با استفاده از مهار کننده آنزیم سازنده نیتریک اکساید (L-NAME) نقش نیتریک اکساید در یادگیری احترازی غیر فعال بررسی گردید. روشها: به این منظور از موشهای صحرایی نژاد ویستار به وزن 200 تا 250 گرم استفاده شد. بوسیله جراحی استرئوتاکسیک، در ناحیه CA1 هیپوکامپ به صورت دوطرفه کانول گذاری انجام شد. یک هفته پس از جراحی آزمایشهای رفتاری صورت گرفت. 25 دقیقه قبل از شروع آموزش در دستگاه یادگیری احترازی غیر فعال، L-NAME در دوزهای 5، 10 و 15 میکروگرم در نیم میکرو لیتر حلال (سالین) در ناحیه CA1 تزریق شد. تست به یادآوری با سه فاصله زمانی بعد از آموزش انجام گرفت. حافظه جاری (بلافاصله بعد از آموزش)، حافظه کوتاه مدت (90 دقیقه بعد از آموزش) و حافظه بلند مدت (24 ساعت بعد از آموزش) ارزیابی شد. یافته ها: نتایج این تحقیق نشان می دهد که تزریق دوز 15 میکروگرم در نیم میکرولیتر L-NAME قبل از آموزش باعث افزایش معنی دار تعداد ورود حیوان به اتاق تاریک در بررسی حافظه جاری و کاهش معنی دار زمان ورود به اتاق تاریک در آزمایش حافظه کوتاه مدت نسبت به گروه حلال شده است. نتیجه گیری: این یافته ها بیان می دارد که مهار ساخته شدن نیتریک اکساید هیپوکامپی قبل از آموزش باعث تخریب حافظه جاری و کوتاه مدت می شود اما اثر معنی داری بر حافظه بلند مدت ندارد. لذا پیشنهاد می شود که احتمالا نیتریک اکساید به عنوان یک پیامبر برگشتی در مراحل ابتدایی یادگیری و حافظه احترازی غیر فعال دخیل می باشد. http://ppj.phypha.ir/article-1-495-fa.pdf 2014-05-15 1 9 hippocampus Nitric oxide passive avoidance learning L-NAME Rat The role of hippocampal nitric oxide in passive avoidance learning Abstract: Introduction: Nitric oxide (NO) is a retrograde messenger in hippocampal synaptic plasticity which involves in learning and memory processes. Previous studies revealed that hippocampal pyramidal cells contain NO synthase (NOS) enzyme which produce NO and could be a promising target to evaluate the role of NO in brain cognitive functions. So in this study, using NOS inhibitor (L-NAME), we conducted an experiment to assess the role of NO in passive avoidance learning. Methods: For this purpose, adult male Wistar rats (200-250 gr) were bilaterally implanted into the CA1 region of hippocampus. A week after surgery animals subjected to behavioral tests. 25 min. before training, rats received different doses of L-NAME (5, 10 and 15 µg/0.5µl/side) into the CA1 of hippocampus. Retrieval tests were performed in three different stages after training as working or immediate memory (immediately after training), short-term memory (90 min. after training) and long-term memory (24 h after training). Results: Our finding showed that pre-training injection of 15 µg/0.5µl/side L-NAME significantly increased the number of step-through into dark chamber and decreased step-through latency, for immediate and short-term memory, respectively. Conclusion: These results suggested that hippocampal NOS inhibition impairs both immediate and short-term memory, but have no any significant effect on long-term memory. OTHERS_CITABLE Role of matrix metalloproteinase II on analgesic effect of nitric oxide inhibition in rat Abstract Introduction: Matrix metalloproteinase 2 is one of the inflammatory mediators that is involved in nociceptive processing and its production is regulated by many inflammatory factors such as nitric oxide. We studied the role of MMP-2 on the analgesic effects of nNOS inhibitor. Methods: Considering that nitric oxide has many roles in pain processing, we studied the CSF levels of MMP-2 after hind paw formalin injection (50µl, 2.5%) and neuronal nitric oxide synthase inhibition intrathecally. We also studied the effect of MMP-2 inhibitor on pain behavior and its role on analgesic effect of neuronal nitric oxide synthase (nNOS) inhibitor. Results: Rats that received MMP-2 inhibitor (30mM) showed severe responses to the formalin injection. Pain was reduced after nNOS inhibition. Prior to nNOS inhibitor injection, MMP-2 inhibition reduced the analgesic effects of nNOS inhibitor. Immunological study showed that MMP-2 increased in rats that received nNOS inhibitor. OTHERS_CITABLE بررسی اثر پیش شرطی سازی هیپرکسی نورموباریک بر فعالیت سوپراکسید دیسموتاز و نارسایی های نورولوژیک در مدل بیماری هانتینگتون مقدمه: مطالعات اخیر بیان می کند که هیپرکسی نورموباریک(HO) باعث ایجاد پدیده تحمل به اکسیتوتوکسیسیتی و استرس اکسیداتیو در انواع اندام ها به خصوص مغز می شود. هدف از این مطالعه بررسی اثر پیش شرطی سازی هیپرکسی نورموباریک بر نارسایی های نورولوژیک و فعالیت آنزیم سوپراکسید دیسموتاز بافت مغزدر مدل جانوری بیماری هانتینگتون است. روش ها: . در این آزمایش رت ها در سه گروه دسته بندی می شوند. جانوران در گروه اول اکسیژن بالای 90 درصد به مدت 4 ساعت در روز به مدت 6 روز استنشاق می کردند. جانوران گروه دوم و سوم هوای اتاق (RA) استنشاق می کردند. بلافاصله بعد از پایان پیش شرطی سازی، مدل سازی بیماری هانتینگتون با تزریق داخل صفاقی روزانه 20 میلی گرم در کیلو گرم وزن بدن اسید نیتروپروپیونیک (3-NP) در شش روز انجام می شد. یافته ها: پیش شرطی سازی با HO ) باعث ایجاد پدیده تحمل به اکسیتوتوکسیسیتی می شود. پیش شرطی سازی با HO در مقایسه با هوای اتاق باعث کاهش معنی دار نقص نورولوژیک مانند تعادل بر روی میله، زمان عقب کشیدن اندام حرکتی، زمان پنجه گیری، افتادن از سطح شیب دار می شد. همچنین فعالیت آنزیم سوپراکسید دیسموتاز در شرایط هیپرکسی افزایش می یافت. نتیجه گیری: اگرچه مطالعات بیشتری برای وضوح مکانیسم های بهبود دهنده پیش شرطی سازی لازم است، اما HO ظاهراً تا حدی آثارشان را از طریق مقاوم سازی سلولها با پیش شرطی سازی و افزایش میزان فعالیت آنزیم سوپراکسید انجام می دهد. http://ppj.phypha.ir/article-1-521-fa.pdf 2014-05-15 18 27 هیپرکسی نورموباریک، هانتینگتون، پیش شرطی سازی، نارسایی های نورولوژیک The Effect of Normobaric Hyperoxia on Superoxide Dismutase Activity and Neurologic Deficits in Huntington Animal Model Introduction: resent studies have been shown that normobaric hyperoxia (HO) can induce excitotoxicity and stress oxidative tolerance (ETT) in variety of organs such as brain. In this study, we examined the intermittent dose of normobaric hyperoxia (HO) on neurologic deficit, and superoxide dismutase activity in brain tissue of Huntington animal model. Method: The rats were divided to three groups. First group were exposed with HO intermittently (4h×6days HO) and second two main group acted as controls, and were exposed to 21% oxygen in the same chamber (room air, RA) discontinuously (4h×6days InRA). Immediately, HO and RA groups were subjected to Huntington animal model (20mg/kg of 3-nitropropionic acid for 6 days), Because Huntington disease is a neurodegenerative disorder. Then, ETT induced by HO were measured by neurologic deficits such as string, limb withdrawal, inclined plane, beam balance test, and assessment of superoxide dismutase activity. Result: Our findings indicated that HO is involved in the induction of ETT. Pretreatment with HO improved neurologic deficits including string, limb withdrawal, inclined plane, beam balance test significantly. Preconditioning with HO increased superoxide dismutase activity significantly. OTHERS_CITABLE نقش مسیر آهسته بعد از سوزاندن مسیر سریع گره دهلیزی بطنی در خواص پایه و وابسته به سرعت گره دهلیزی بطنی جدا شده خرگوش   مقدمه : هدف از این مطالعه کسب دید تازهای در ارتباط احتمالی بین خواص کارکردی مسیر آهسته پنهان و خواص وابسته به سرعت گره دهلیزی بطنی است.   روش ها : در یک گروه شامل 7 نمونه از گره دهلیزی بطنی با تغذیه سطحی خرگوش پروتکل های تحریکی وابسته به سرعت ریکاوری ، تسهیل و خستگی اجرا شدند. سوزاندن با کمک ولتاژ مستقیم (110-100 ولت) توسط الکترود نقره تک قطبی انجام گرفت.   یافته ها : سوزاندن مسیر سریع باعث کاهش معنی دار میزان تسهیل شد ولی روی زمان خستگی و تحریک ناپذیری گره ای تاثیر ی نداشت. مهمترین اثر سوزاندن مسیر سریع افزایش معنی دار زمان هدایت حداقل بود.   نتیجه گیری : سوزاندن مسیر سریع سبب آشکار شدن اثرات مسیر آهسته پنهان می شود. خواص وابسته به سرعت گره دهلیزی-بطنی به تداخل دینامیک دو مسیر آهسته و آهسته پنهان بستگی دارند. مسیر سریع در ایجاد پدیده تسهیل موثر است. http://ppj.phypha.ir/article-1-491-fa.pdf 2014-05-15 28 36 سوزاندن ، مسیر سریع ، گره دهلیزی – بطنی، آریتمی Role of slow pathway after nodal fast pathway ablation on the basic and rate- dependent properties of the isolated rabbit atrioventricularNode   Introduction : The aim of this study is to obtain new insight into possible relation between functional properties of slow concealed pathway and rate-dependent properties of AV-node.   Methods : Rate-dependent nodal properties of recovery, facilitation, and fatigue were characterized by stimulation protocols in one groups of (N=7) isolated superfused AV-Nodal rabbits. Small miniature lesions were made by delivering constant voltage (110 V-100 s) with unipolar silver electrode.   Results : Fast pathway ablation significantly decreased facilitation and had no effect on fatigue and nodal refractoriness. The most important effect of fast pathway ablation was prolongation of minimum conduction time.   Conclusion : The FP-ablation revealed the presence of the concealed SP. Rate-dependent property of node is dependent to dynamic interaction between concealed slow with slow pathway. OTHERS_CITABLE القای هایپواینسولینمیا و تاثیر آن بر ترشح لپتین، متابولیت‌های خون و ادرار، مقدمه: نقش و اهمیت انسولین در نشخوارکنندگان متفاوت از تک‌معده‌ایهاست. در این تحقیق هایپواینسولینمیا دائمی با شدت‌های متفاوت توسط دزهای 25 (کم)، 50 (متوسط) و 75 (بالا) استرپتوزوتوسین (mg/kg BW) در گوسفند ایجاد شد. روش‌ها: بیست بره پرواری به چهار گروه تیماری تقسیم و بصورت انفرادی نگهداری شدند. تیمارها شامل شاهد و تزریق منفرد و درون‌رگی سه دز استرپتوزوتوسین بود. دوره تحقیق شامل هشت هفته متوالی بود که تزریق استرپتوزوتوسین در پایان هفته سوم انجام گرفت. نمونه‌های خون از رگ وداج به طور هفتگی و در حالت‌های ناشتا و 5/2 ساعت بعد تغذیه جمع‌آوری و آنالیز شد. مصرف خوراک و آب و همچنین تغییرات وزن حیوانات اندازه‌گیری گردید. بعد از کشتار، اندام‌های درونی توزین شد و نمونه ادرار از مثانه جمع‌آوری گردید. یافته‌ها: با تزریق دز بالا، حیوانات دچار شرایط غیرعادی شده و نتوانستند آزمایش را ادامه دهند. نتایج حاکی از بروز هایپواینسولینمیا با تزریق دز متوسط بود که با کاهش معنی‌دار غلظت انسولین ناشتا و بعد تغذیه و همچنین غلظت لپتین ناشتا نسبت به شاهد همراه بود (05/0>P). دز متوسط باعث افزایش معنی‌داری در سطح گلوکز، تری‌گلیسریدها، کلسترول، پروتئین تام، ازت اوره‌ای و همچنین اجسام کتونی خون نسبت به شاهد شد (05/0>P). حیوانات با تزریق دز متوسط، پرخوری دیابتی و افزایش مصرف آب را در هفته‌های بعد تزریق نسبت به شاهد نشان دادند (05/0>P) ولی علیرغم مصرف غذای بیشتر نتوانستند وزن بیشتری نسبت به شاهد کسب کنند. غلظت قند و پروتئین ادرار به طور وابسته به دز افزایش داشت ولی معنی‌دار نبود (05/0<P). وزن و شاخص اندام‌های داخلی بدن بین گروه‌ها تفاوت معنی‌داری نشان نداد فقط وزن لاشه در گروه دز متوسط نسبت به دو گروه دیگر بیشتر بود. نتیجه‌گیری: نتایج بیانگر نقش حیاتی تنظیمی انسولین در متابولیزم انرژی نشخوارکنندگان بوده که با اعمال دو اثر متضاد کاتابولیک مغزی و آنابولیک محیطی می‌باشد که موافق با یافته‌ها در تک‌معده‌ایها بود. http://ppj.phypha.ir/article-1-515-fa.pdf 2014-05-15 37 47 هایپواینسولینمیا، لپتین، استرپتوزوتوسین، متابولیت‌های خون، گوسفند Hypoinsulinemia induction and its effects on leptin secretion, blood and urine metabolites, feeding pattern and internal organs indices in sheep Introduction: Insulin role and importance are different from that of monogastric. In this study, permanent hypoinsulinemia with various severities was induced using doses 0, 25, 50 and 75 mg/kg BW of streptozotocin (STZ) in sheep. Materials and methods: Twenty male lambs were divided into four treatment groups and maintained individually. Treatments were single, intravenous injection of doses of STZ. The duration of the experiment was eight consecutive weeks, and injection was performed at the end of third week. Blood samples were collected weekly via venipuncture at fasting and 2.5 h post-prandial times and analyzed. Feed and water intakes and also animals' weight changes were measured weekly. After slaughter, internal organs were weighed and urine samples were collected from bladder. Results: Animals receiving dose 75 could not continue the experiment because of abnormalities. Results showed the occurrence of hypoinsulinemia with injection of dose 50 with significant decrease in fasted and post-prandial insulin concentrations and also fasted leptin concentrations vs dose 0 as control (P<0.05). Dose 50 caused significant increase in blood glucose, triglycerides, cholesterol, total protein, blood urea nitrogen and keton bodies levels vs control (P<0.05). Animals receiving dose 50 showed diabetic hyperphagia and enhanced water intake in weeks after injection as compared to control (P0.05). Weights and indices of internal organs showed no differences between groups, but only carcass weight in group of dose 50 was significantly greater than others. OTHERS_CITABLE متوقف شدن روند آندوسیتوز کانال پتاسیمی ROMK2 (Kir1.1b) در اثر موتاسیون S362A درغشاء اووسیت های Xenopus laevis متوقف شدن روند آندوسیتوز کانال پتاسیمی ROMK2 (Kir1.1b) در اثر موتاسیون S362A درغشاء اووسیت های Xenopus laevis سعید حاجی هاشمی1 1- استادیار، دکتری تخصصی فیزیولوژی ،عضو هیئت علمی ، گروه فیزیولوژی ،دانشکده پزشکی، دانشگاه علوم پزشکی اراک چکیده: مقدمه : کانال های پتاسیمی ROMK در غشاء راسی سلولهای کلیه قرار گرفته اند. ایزو فورم های مختلفی از کانال پتاسیمی ROMK در قسمت های انتهای نفرون و در مجاری جمع کننده شناسایی شده اند که وظیفه ترشح یون K+ را بر عهده دارند. تحقیقات نشان داده اند که آندوسیتوز کانالهای پتاسیمی ROMK برای ترشح K+ در مجاری جمع کننده نقش مهمی دارا می باشد. در این مطالعه اثرات موتاسیون S362A (جایگزین کردن اسید آمینه سرین در جایگاه شماره 362با اسید امینه آلانین) بر روی اندوسیتوزکانال پتاسیمی ROMK2 پس از بیان درغشاء اووسیت بررسی گردیده است. روش کار:در این پ‍ژ‍وهش تجربی اووسیت های Xenopus laevis با استفاده از کلاژناز به روش استاندارد جدا گردیدند و جهت ایجاد موتاسیون در انتهای کربوکسیل کانال پتاسیمی ROMK2 با استفاده از روش ایجادتغییر سریع ایجاد موتاسیون زای مستقیم گردید. cRNA ای کهROMK2 وموتاسیون S362A را کد می کرد به اووسیت ها تزریق شد. پس از گذشت سه روز( زمان صفر) به محیط کشت برفلدین A (+BFA) مهار کننده انتقال پروتئینهای ساخته شده به غشاء به مقدار 25 میکرومولار یا اتانول به عنوان حلال BFA(BFA -) اضافه گردید. با استفاده از تکنیک ثابت نگه داشتن ولتاژ با استفاده از دو الکترود جریانهای یونی مربوط به کانال های پتاسیمی ROMK2 و موتاسیون S362A اندازه گیری گردید. نتایج: یافته ها نشان داد که مقدار جریان یونی پتاسیم از کانالهای پتاسیمی ROMK2با موتاسیون S362A بر خلاف کانال های پتاسیمی بدون موتاسیونROMK2 پس از انکوبه شدن در محلول 25 میکرو مولار BFA کاهش معنی داری پیدا نکرد. برای ROMK2 پس از اینکه اووسیت ها 48 ساعت تحت تاثیر BFA بودند میزان کسر جریان برابر با 0.24  0.05 (n=16)بود. این نتایج افزایش پایداری وثبات کانالهای پتاسیمی ROMK2 درغشاء بعدازایجاد موتاسیون S362A را نشان می دهد نتیجه گیری: قسمت داخلی ناحیه PDZ به ترتیب با اسید های سرین – گلوتامیک اسید - والین ( S-E-V) در تعیین پایداری و آندوسیتوز کانالهای پتاسیمیROMK2 در غشاء سلول خالت دارد. کلید واژگان: ROMK2، موتاسیون S362A ، BFA ، ناحیه PDZ نویسنده مسئول: اراک ،میدان بسیج ، مجتمع دانشگاه علوم پزشکی اراک ، دانشکده پزشکی ،گروه فیزیولوژی Email:hajihashemi@hotmail.com http://ppj.phypha.ir/article-1-478-fa.pdf 2014-05-15 48 56 ROMK2، موتاسیون S362A ، BFA ، ناحیه PDZ The S362A mutation block ROMK2 (Kir1.1b) endocytosis in Xenopus laevis oocyte membrane . Introduction: ROMK channel is localized on the apical membrane of the nephron. Recent studies suggest that endocytosis of ROMK channels is important for regulation of K+ secretion in cortical collecting ducts. In this study the effect of S362A mutation is examined on the membrane turnover and stability of ROMK2 channel when expressing in Xenopus laevis oocytes. Materials and Methods: In this experimental study oocytes were isolated by standard protocols using collagenase (Type 1A). Mutations of the cytoplasmic termini of ROMK2 were constructed using the quik-change approach for site-directed mutagensis. Xenopus oocytes were injected with cRNA encoding ROMK2 or S362A mutant three days prior to treatment with BFA solution (time 0). Brefeldin A (BFA) was added to the OR3 medium (+BFA) at concentrations of 25µM (inhibit insertion of new proteins into the cell membrane) or ethanol as BFA vehicle (-BFA). Two-electrode voltage clamp (TEVC) was used to measure oocyte ROMK-dependent currents and membrane potential. Data was analyzed using Student’s t-tests or ANOVA as appropriate. Results: Incubation of oocytes expressing ROMK2 channels in 25 μM BFA caused a reduction in the currents and membrane voltage. In oocytes expressing the S362A mutant, there was no decay in current and membrane voltage after 48 hours incubation with BFA at 25 µM. The fractional current for ROMK2 at 48h following treatment of oocytes with BFA was 0.24 0.05 (n=24) which was significantly different to S362A mutant (0.96 0.05 n=24). Conclusion: These results show that the S362A mutation increases the general stability of ROMK and renderes the protein resistant to endocytosis, consistent with the idea that there is an interaction between the C-terminal of ROMK2 and components of the endocytotic pathway. OTHERS_CITABLE تیمار با BIO سبب افزایش میزان تکثیر و توان زیستی سلول های بنیادی مزانشیمی مشتق از مغز استخوان موش صحرایی در محیط کشت می شود   مقدمه: بر اساس مطالعات پیشین، حضور BIO ( (6-Bromoindirubin-3΄-Oxime در محیط کشت برخی سلول ها سبب افزایش بقا وتکثیر آن می شود. هدف تحقیق حاضربررسی اثرات این ماده بر تکثیر سلو ل های بنیادی مزانشیمی مشتق از مغز استخوان موش صحرایی است. روش ها: در این مطالعه تجربی، سلول های مغز استخوان 7 سر موش صحرایی در حضور غلظت های 01/0 ، 05/0، 1/0، 1 و 5/1 میکرومولار BIO کشت ش د و با انجام سه پاساژ متوالی تکثیر گردید. کشت سلول در گروههای مختلف، در طول مدت کشت از لحاظ برخی شاخص های رشد شامل قطر و تعداد کلونی های تشکیل شده ، میزان دو برابر شدگی جمعیت سلولی و تعداد سلول های زنده مقایسه شد. در پایان سلول های پاساژ 3 در گروههای مختلف از لحاظ حفظ توان تمایز به استخوان و چربی ارزیابی شد. یافته ها: بر اساس نتایج، بزرگترین کلونیها در گروههای 1/ 0 و1 میکرومولار و به ترتیب با قطر 96/43 ± 27/1262 و16/19 ± 71/1335 میکرومترو تشکیل شد. از لحاظ تعداد کلونی نیز این دو گروه به گروه های آزمایشی دیگر و کنترل برتری داشتند ( P<0.05 ) . در طی 3 پاساژ متوالی درگروه 1/0 و1میکرومولار جمعیت سلولی نسبت به گروه های آزمایشی و کنترل دو برابر شد ( P<0.05 ) . همچنین، در این دو گروه ، تعداد سلول های زنده بیش از گروه های دیگر بود ( P<0.05 ) . رنگ آمیزی آلیزارین رد برای استخوان و اویل رد برای چربی نشان داد که سلول های گروههای مختلف توان تمایزی خود را حفظ کرده اند نتیجه گیری: در مجموع می توان گفت که تیمار کشت سلول های مغز استخوان موش صحرایی با BIO بویژه با غلظت های 1 و 1/0 میکرومولار سبب افزایش سرعت رشد و نیز توان زیستی سلول ها می شود. http://ppj.phypha.ir/article-1-484-fa.pdf 2014-05-15 57 67 سلول بنیادی مزانشیمی، موش صحرایی، BIO ، تکثیر BIO treatment enhances rat marrow-derived mesenchymal stem cell in vitro proliferation and viability Introduction: Previous investigations have indicated that the presence of BIO (6-Bromoindirubin-3-Oxime) in medium of some cell culture enhances the cell proliferation and viability. The aim of the present study was to investigate the BIO effects on in vitro expansion of rat marrow-derived mesenchymal stem cells (MSCs) culture. Methods: In the present experimental study, bone marrow cells from 7 rats were plated in the presence of 0.05, 0.01, 0.1, 1 and 1.5 µM of BIO and expanded through three successive subcultures. During the cultivation period, the cultures were statistically compared in terms of some indices of cell growth including the diameter and number of colonies, population doubling number (PDN) and the number of viable cells. Passaged-3 cells from all groups were examined whether or not they could differentiate into bone and adipose cells. Results: According to our results, the largest colonies were formed in the cultures with 0.1 and 1 µm BIO with diameter of respectively 1262.27±43.96 and 1335.71 ± 19.16 micrometer (P<0.05). These two groups were also superior in terms of the colonies numbers. During three successive passages, significantly more PDN was occurred in 0.1 and 1 µM BIO- treated cultures than the others (P<0.05). Additionally in these two BIO-treated cultures, the number of viable cells was significantly higher compared to other BIO-treated cultures as well as the control group (P<0.05). OTHERS_CITABLE Effect of esophagus distension on gastric blood flow, gastrin and somatostatin secretion in rat Abstract Introduction: There are many studies about the inhibitory effect of the esophageal distention (ED) on gastric motility. Recently, it has been shown that ED decreases the gastric secretions. It is well established that the inhibitory effect of ED is mediated by activation of vago-vagal inhibitory reflex. However, there is not any investigation about the effect of the reflex on the gastric blood flow and release of gastric hormones. The aim of this study was to investigate the effect of esophageal distention (ED) on gastric blood flow and release of gastrin and somatostatin hormones. Methods: In this study 79 male Wistar rats (175-230 g) were used. deprived of food but not water 24 h before the experiments. Under urethane anesthesia (1.2 g/kg, i.p.), animals underwent tracheostomy and laparotomy. A catheter was inserted in the stomach through duodenum for gastric distension. The esophagus was cannulated with a balloon orally to distend distal portion of esophagus (0.3 ml, 10 min). Saline was used for gastric distension (1.5 ml/100 g, b.w., pH 7 and 37 °C). Gastric blood flow was measured by a Laser Doppler flowmeter. Gastrin serum and somatostatin plasma levels were assessed by RIA method. Vagotomy was carried out in a group to reveal the role of vagus nerve in this action. Results: ED reduced the blood flow of gastric proximal portion (P<0.001) but the antrum blood flow was unaffected by ED. Cervical vagotomy abolished the inhibitory effect of the ED on the proximal area blood flow. Gastrin serum level and somatostatin plasma level were unaffected by ED. Conclusion: Removal of the inhibitory effect of ED after vagotomy shows that the vagus nerve was involved in the inhibitory effect on gastric blood flow. OTHERS_CITABLE The comparison of normal venous blood with   Introduction: Cupping therapy (hejamat) is one of the therapeutic methods used in traditional medicine that it is still common in many of the contemporary societies. In spite of the progress of science, we have less information on the traditional and alternative medicine including cupping.   With the study of mechanism of action of cupping, it will be more convincing for the traditional medicine to use it more easily for the treatment. It can then be presented to society with more convincing scientific base.   The aim of this study was to compare venous blood with blood of cupping with regards to biochemical, hematological and immunological factors and evaluate the variation of those factors.   Materials: The sample of study was 56 healthy volunteer men aged between 20 to 40 years. At the beginning of study (the first) 16 ml blood sample from vein was obtained from each volunteer for the comparison of biochemical, hematological and immunological factors. The cupping treatment was then given to the volunteers and another blood sample from cupping was taken for comparison of these factors.   The biochemical factors were evaluated by Selectra auto analyzer, hematological factors by KX-21 cell counter and sedimentation rate was measured by Westergreen method. Cytokines were measured by sensitive sandwich ELISA, using the kit from R;D system.   Results: Analysis of the results showed significant differences in many of the biochemical, hematological and immunological factors between the venous blood and the cupping blood. OTHERS_CITABLE Effect of intracerebroventicular injection of baclofen and CGP35348 on pain sensitivity during estrous cycle Introduction: In female rats sensitivity to antinociceptive treatment varies during estrous cycle stages. The role of GABA through GABAB receptor in nociception has been established. The aim of the present study is to investigate the effect of intercerebroventicular injection of GABAB receptor agonist (baclofen) and GABAB antagonist (CGP35348) on pain sensitivity during different stages of estrous cycle. Methods: Forty adult female rats weighing 200-220g were used. Rats were maintained on 12 h reversed light/dark cycle, and standard temperature 22 ± 2°C. Food and water were available ad libitum. Pain sensitivity was evaluated by formalin test, performing by subcutaneous injection of 50µl formalin solution (2.5%) into the hind paw in each stages of estrous cycle. Data were analyzed by two ways ANOVA measuring and Tucky test as post-hoc test. The level of significance was P<0.05. Results: Our data showed that baclofen significantly decreased pain sensitivity in all stages of estrous cycle (P<0.05). The analgesic effect of baclofen was significantly higher during metestruse and diestruse as compared to proestruse and estrus (P<0.05). CGP35348 significantly increased pain sensitivity in all stages of estrous cycle (P<0.05). The hyperalgesic effect of CGP35348 was significantly lower during metestruse and diestruse than proestruse and estrus (P0.05). The effect of aerobic exercise on coagulation and fibrinolysis has been studied extensively Regarding to the effectivness of resistance exercises to increase functional ability of men, the purpose of this study was to examine the effects of the combined aerobic-resistance and aerobic exercise on the coagulation activity of healthy young men. 30 young healthy sedentary men (20±5 years old) randomly divided in 3 groups: 10 in the combined, 10 in the aerobic and 10 in nontraining matched control groups The training groups subjects were engaged in a program included, ten session, three times per week, with submaximal intensity and 24 min for every session. The combined group had 12 min resistance exercise, followed by a 12 min aerobic exercise on cycle ergometer although aerobic group had only 24 min aerobic exercise on cycle ergometer. The results revealed that PT decreased and PTT increased in training groups. Fibrinogen decreased significantly in both training groups.
