نقش پروتئین S100B آستروسیتی بر تقویت حافظه در موشصحرایی

مقدمه : پروتئین S100B ، پروتئینی است که از آستروسیت ها آزاد می شود و در کنترل ارتباط نوروگلیالی و احتمالا در حافظه و شکل پذیری سیناپسی نقش دارد. مطالعه ارتباط عملکردی دو سلول آستروسیت و نورون از اهمیت خاصی برخوردار است. در مطالعه حاضر اثر تزریق این پروتئین به درون هیپوکامپ بر روی یادگیری در موش های صحرایی مورد بررسی قرار گرفت.
روش ها: در این تحقیق از 40 موش صحرایی نر استفاده شد. حیوانات پس از جراحی و کانول گذاری دو طرفه در ناحیه CA1 هیپوکامپ و نیز 7روز ریکاوری ، تحت یادگیری احترازی غیرفعال قرار گرفتند (5/0 میلی آمپر, 100 هرتز, 5 ثانیه). بلافاصله پس از آموزش، 5/0 میکرولیتر پروتئین S100B , به صورت دوطرفه ,در دوزهای مختلف (5 و 50 و 500 و 5000 نانوگرم) به گروه های آزمایشی و سالین به گروه کنترل تزریق شد . شاخص های یادگیری و حافظه شامل اولین تاخیر در پایین آمدن از سکو و کل زمان سپری شده بر روی سکو ، 24 ساعت بعد سنجش شد.
یافته ها: تزریق5 نانوگرم S100B ، افزایش معنی داری را در شاخص اولین تاخیر در پایین آمدن از سکو (01/0< P ) و نیز کل زمان سپری شده بر روی سکو (001/0< P) نشان داد و بطور شگفت آوری دوزهای 500 و 5000 نانوگرم این پروتئین ، درمقایسه با گروه کنترل کاهش معنی داری را در هر دو شاخص فوق الذکر نشان دادند (001/0< P) (001/0< P).
نتیجه گیری: یافته های ما نشان می دهد پروتئین آستروسیتیS100B ، اثر ات تعدیلی بر روی حافظه دارد، بطوریکه در دوز های نانوگرم حافظه را تسهیل ، در حالیکه در دوزهای میکروگرم حافظه احترازی غیر فعال را تخریب می کند.
Introduction: S100B is a protein released from astrocytes, which controls neuroglial relationship and probably
plays a role in memory and synaptic plasticity. Study of the functional relationship between astrocytes and neurons is of
great importance. The present study was conducted to evaluate the effect of S100B infusion into the CA1 region of the
hippocampus, on memory performance in male rats.
Methods: In this study, 40 male wistar rats were used. Animals were bilaterally implanted with indwelling cannulae
in the CA1 region of the hippocampus. Seven days after surgery, animals were trained in a step-down passive avoidance
task (0.5 mA, 100 Hz, 5 sec). Immediately after the training, animals received 0.5 μl infusion of saline or S100B (5, 50,
500, or 5000 ng) bilaterally. Twenty four hours later step-down first latency and total time spent on platform were
measured as learning and memory indices.
Results: The infusion of 5 ng S100B induced a significant increase in step-down first latency (p< 0.01), and also
increased the total time spent on the platform compared to the control group (p < 0.001). Surprisingly, animals which
received doses of 500 and 5000 ng showed a significant decrease in both indices compared to the control group (p<
0.001).
Conclusion: Our findings indicate that astrocytic S100B protein has modulatory effects on memory, in a way that in
nanogram doses facilitates, but in micrograms impairs memory in passive avoidance task.
